Fungal mutants not solved by internal amplification controls.

چکیده

I was interested to read Rodríguez et al. (2012) on PCR to detect verrucosin producing fungi in food and cultures. However, their interpretation regarding mutated strains and internal amplification controls (IAC) appears to be incorrect, although they referred to our papers on the issue. It is worth restating to the wider scientific community that fungi may be mutated by self produced mutagens and enzyme inhibitors leading to false negative or positive results (Paterson et al., 2008; Paterson and Lima 2009) for which an IAC is irrelevant. Finally, interpretations of PCR results from cultures or food require to consider these possibilities. I look forward to your reply.

Biocontrol Activities of Gamma Induced Mutants of Trichoderma harzianum against some Soilborne Fungal Pathogens and their DNA Fingerprinting

Background: Random induced mutation by gamma radiation is one of the genetic manipulation strategies to improve the antagonistic ability of biocontrol agents. Objectives: This study aimed to induce mutants with more sporulation, colonization rate leading to enhanced antagonistic ability (in vitro assay) comparing to wild type (WT) and the assessment of genetic differences (in situ evaluat...

Internal amplification controls have not been employed in fungal PCR hence potential false negative results.

Polymerase chain reaction (PCR) is subject to false negative results. Samples of fungi with the genes of interest (e.g. a disease or mycotoxin) may be categorized as negative and safe as a consequence. Fungi are eukaryotic organisms that are involved in many fields of human activity such as antibiotic, toxin and food production. Certain taxa are implicated in human, animal and plant diseases. H...

Nutrient enrichment increased species richness of leaf litter fungal assemblages in a tropical forest.

Microbial communities play a major role in terrestrial ecosystem functioning, but the determinates of their diversity and functional interactions are not well known. In this study, we explored leaf litter fungal diversity in a diverse Panama lowland tropical forest in which a replicated factorial N, P, K and micronutrient fertilization experiment of 40 × 40 m plots had been ongoing for nine yea...

Molecular probes for the detection of pathogenic fungi in the presence of human tissue.

Four primer systems, amplifying fragments of the gene coding for the small ribosomal subunit (18S rRNA) were characterised with pure cultures of 65 medically relevant fungal species plus two mushrooms. A primer cocktail (TR1/CA1-TR2/AF2) amplified 59 of 67 fungal species; the universal fungal primer 1 (UF1) in combination with the eukaryotic primers S3 or EU1 amplified 64 and 65 of 67 fungal sp...
